IM-1021
(ROR1 Antibody-Drug Conjugate)

ROR1 is a receptor that is expressed on liquid and solid tumors but is largely absent in most mature tissues. IM-1021 is a ROR1-targeted ADC that is optimized for ADC-mediated killing of ROR1 positive tumor cells. It incorporates an antibody designed to promote internalization and uses a cleavable undisclosed linker – a proprietary camptothecin derivative payload, HC74 – designed to maximize the potential bystander effect and support a drug-antibody ratio of 8.

We believe that IM-1021’s combination of attributes could result in an improved therapeutic index when compared to other ROR1-targeted ADCs currently in development. In preclinical studies, IM-1021 showed marked tumor regression in mouse models of triple-negative breast cancer, non small cell lung cancer, and mantle-cell lymphoma. We initiated enrollment in a Phase 1 study of IM-1021 in the first quarter of 2025.
